What Ginsenoside Rg3 is
A ginseng saponin whose effect on senescence runs in both directions depending on the cell: it reversed replicative senescence in human fibroblasts in culture and is studied as a senescence inducer in tumour cells. The only human data are pharmacokinetic.
Ginsenoside Rg3 is a triterpenoid saponin from Panax ginseng, studied mainly in China and Korea. Its senescence literature is cell culture.
Mechanism: In cultured human fibroblasts, 20(S)-Rg3 restored ATP and the NAD+ to NADH ratio, downregulated PI3K and Akt through mTOR inhibition with p53 and p21 involvement, and activated sirtuin 3 and PGC1alpha to stimulate mitochondrial biogenesis; the 20(R) stereoisomer did not. In astrocyte culture, Rg3 suppressed interleukin 6 and interleukin 8 via NF-kappaB and p38 MAPK, a senomorphic action.
Regulatory history: Not approved for any indication in the United States. Sold as a ginseng-derived supplement ingredient. No senescence or ageing trial is registered.
How MHRA treats this kind of product
Licenses medicines for the UK market. A compound without a marketing authorisation is an unlicensed medicine, which can only lawfully be supplied for human use through a prescriber under the specials scheme, not sold as a research product.
The classification above is the The Longevity Archive reading of Medicines and Healthcare products Regulatory Agency listings for Ginsenoside Rg3 at the reviewed date, not a legal opinion. Rules change, and importing, prescribing and possessing a compound can each be treated differently. Check the regulator directly before acting on anything here.
