A commercial supplement that genuinely extended male mouse lifespan in the government testing programme, which almost no supplement can claim. Median only, males only, and no human ageing data.

Overview

Protandim is unusual on this site: a product sold through multi-level marketing that was independently tested by the National Institute on Aging and produced a real result.

In the Interventions Testing Program it extended median lifespan in male mice. That is a genuine finding from the most rigorous lifespan testing apparatus that exists, obtained without the company controlling the study.

The honest framing requires the rest of the sentence. Median lifespan only, not maximal. Males only, not females. Mice, not people. And no human trial has examined lifespan, healthspan or any ageing endpoint.

It is a mixture of five botanical extracts that activate Nrf2, the transcription factor controlling the antioxidant response, which places it in the same mechanistic family as the dimethyl fumarate entry on this site whose lifespan test was compromised by underdosing.

Mechanism of action

A combination of botanical extracts including milk thistle, bacopa, ashwagandha, green tea and turmeric, selected to activate nuclear factor erythroid 2-related factor 2. Nrf2 is held inactive by KEAP1 and released under electrophilic stress, whereupon it drives transcription of antioxidant response element genes including glutathione synthesis enzymes and haem oxygenase 1. The proposed logic is indirect: rather than supplying antioxidants, it prompts the cell to make its own, which avoids the problem that direct antioxidant supplementation has repeatedly failed to extend lifespan. Being a mixture, no effect can be attributed to any single component.

What this does not tell you: The result is real and narrow: median lifespan, males only, mice. Because it is a mixture, the finding belongs to that specific combination and formulation rather than to Nrf2 activation in general, and it does not transfer to any other Nrf2 product. The wider Nrf2-and-lifespan question remains open, since the one attempt with a well-characterised pharmaceutical Nrf2 activator delivered only 35 percent of its target dose.

Reading the research record

This page is a test of whether the site can report a result it might prefer not to. Protandim is sold through multi-level marketing, which is exactly the distribution model this site's owner tells people to avoid, and it nonetheless produced a genuine lifespan extension in the most rigorous mouse programme in existence, tested independently by a government agency rather than by its manufacturer.

That deserves reporting accurately, which means reporting all of it: median lifespan, males only, in mice, with no human ageing data, in a mixture where no individual ingredient can be credited.

It is also worth setting beside the dimethyl fumarate entry here. That compound is a well-characterised pharmaceutical Nrf2 activator whose lifespan test delivered an average of 35 percent of the target dose and found nothing, with the authors offering the underdosing as the likely explanation. So the Nrf2 hypothesis currently has one positive from a botanical mixture in male mice and one compromised null from a pharmaceutical. That is not a settled picture in either direction.

The male-specific pattern is the most under-discussed finding in this whole field. Several of the programme's most reliable positives extend lifespan in male mice and not in female mice, and the effect is not a matter of statistical power: the female curves simply do not move. Nobody has established why. Proposed explanations include differences in drug metabolism, in baseline mortality causes between the sexes in this strain, and in hormonal context. What it means practically is that a headline reading a compound extends lifespan in mice is frequently describing an effect in half the animals, and that half is usually not stated.
