GIP is one of the two mammalian incretins. It is released by K cells in the upper small intestine when you eat and it amplifies the insulin your pancreas puts out in response to glucose.

Mechanism: Binds GIPR, a class B1 secretin-family G protein-coupled receptor that is canonically Gs-coupled and raises cAMP. In the pancreatic beta cell that potentiates glucose-stimulated insulin secretion, which is glucose-dependent, so GIP does not drive insulin release when glucose is normal.

Regulatory history: Not an approved drug and not in clinical use as a therapeutic. Drugs acting at its receptor are approved, including tirzepatide.
