This is how I would explain it to a friend. The evidence, with the numbers, is further down the page.
PE-22-28 is a peptide meant to lift depression fast, within days instead of weeks. No human has ever taken it in a study. It is a short piece of a brain protein, cut down to seven building blocks so it lasts longer. It blocks one channel in brain cells that is linked to low mood. It is not approved, and it is sold as a research chemical.
What people take it for
- Fast relief from depression when normal pills have failed or take too long.
- Growing new brain cells.
- Better focus and less brain fog.
- Getting off antidepressants.
What studies in people showed
There are no human trials. The parent peptide, spadin, was given to mice. After only four days, the mice acted less depressed on five different tests. PE-22-28 is a shorter, tougher version. In a dish it blocks the target channel much harder than spadin. In mice it cut the time they gave up and floated in a swim test, which is how scientists measure depression in a mouse. That is the whole record. Nobody knows what it does to a human mood, what a human dose is, or what happens if you take it for a month.

What is PE-22-28?
PE-22-28 is an experimental peptide studied in mice for fast antidepressant effects and new brain cell growth. It blocks a potassium channel in the brain.
PE-22-28 is a shorter lab-made version of spadin. Spadin is a peptide that blocks TREK-1, a potassium channel in the brain linked to depression. No study has tested either one in people. It is experimental and not approved.
In mice, spadin locked onto TREK-1 very tightly (10 nM) and acted like an antidepressant in five behaviour tests after only four days. That is what made people interested in the idea of a fast-acting antidepressant. PE-22-28, just seven amino acids long, blocks TREK-1 far more strongly: it takes 0.12 nM, against 40 to 60 nM for spadin. In mice, it helped in a swim test and in other tests used to model depression.
How does PE-22-28 work?
It blocks TREK-1, a potassium channel tied to mood. In animals, that gave fast antidepressant-like effects.

In mice, the seven amino acid version PE 22-28 blocked TREK-1 far more strongly than spadin. It helped in a swim test and in other tests used to model depression.
The 7-amino-acid analogue PE 22-28 inhibited TREK-1 with an IC50 of 0.12 nM (versus 40 to 60 nM for spadin) and reduced immobility in the forced swim test and other depression models in mice.
