RLS-1496, topical GPX4-modulating senolytic

Rubedo's topical senolytic that met its Phase 1 safety endpoint in psoriasis, eczema and photo-aged skin, with collagen gene expression rising in treated skin.

Overview

RLS-1496 is designed to clear pathologic senescent skin cells by modulating glutathione peroxidase 4. In a Phase 1 reported in March 2026, it was well tolerated; a quarter of psoriasis and atopic dermatitis participants had a 4-point or greater drop in itch at four weeks versus none on vehicle, and photo-aged skin showed dose-dependent target engagement with increasing collagen gene expression. It is the first senolytic with human skin-aging data.

Mechanism of action

Selective modulation of GPX4 in senescent cells, triggering their removal while sparing healthy cells.

Is this the same as senolytic skincare sold online?

No. Cosmetic products marketed as senolytic have no comparable human data. RLS-1496 is a drug candidate in regulated trials.
