This is how I would explain it to a friend. The evidence, with the numbers, is further down the page.

Endoluten is a capsule sold for sleep, hormones and a longer life. It is meant to come from the pineal gland. Search for it in the medical database and you get zero papers. Zero. Every big number used to sell it belongs to two other substances, and neither one is what is in the bottle.

What people take it for

  • Sleeping better and getting the body clock back in order.
  • Living longer, because the marketing leans hard on that.
  • Hormone balance in your 60s and beyond.
  • Feeling sharper and less worn down.

What the trials actually showed

Three names get mixed together here, so keep them apart. Epithalamin is a shot made from cow pineal glands. Epitalon is a lab made peptide built from it. Endoluten is a capsule with no research under its name. All the famous numbers are from epithalamin. In animals it raised average life span by 11 to 31 percent, though in one group of mice it did nothing. In 266 older people followed 6 to 8 years, deaths dropped by about 1.6 to 1.8 times. In a 12 year study of older heart patients, deaths were 28 percent lower. Those are real published numbers. They were all produced by the institute that sells the product, nobody outside has repeated them, and there is no registered trial for anything in this family. A separate pineal shot called Pineamin raised a melatonin marker 1.9 times in 55 older people, with no control group.

What people report

Reports, not trial results

The good

  • Better sleep is the most common report by a wide margin.
  • Vivid dreams in the first week or two.
  • Some people say they wake up at a steadier time.
  • The capsule course is short, so it is easy to try and stop.

The bad

  • Headaches and grogginess in the first few days, reported often on forums.
  • Plenty of people report nothing at all.
  • No safety data exists. Not one study in this family lists side effects, which means nobody looked.
  • Nobody has tested what is in the capsule, so you cannot know if it matches the shot used in those studies.

Where these come from: Sleep and dream reports come from peptide forums and vendor pages. The death and life span numbers come from published Russian studies of the injectable gland extract, not of this capsule.

My bottom line

Those mortality numbers are real papers, and they were written by the people selling the product, with nobody else checking. Buying a capsule and quoting a study done on a cow gland shot is the mistake almost everyone makes here, and I would not spend money on it.

Overview

A pineal capsule sold on the longevity claims of two other substances. A PubMed search for Endoluten returns zero records. Every lifespan and mortality number attached to it was produced with epithalamin, a bovine pineal extract, or with epitalon, a synthetic tetrapeptide, and neither has been shown to be what is in the capsule.

Endoluten is one of the most heavily marketed products in the peptide bioregulator retail line, sold as a pineal gland complex for sleep, hormonal rhythm and longevity. Searched against PubMed the name returns nothing at all. Zero records.

The numbers used to sell it come from two related but distinct substances, and the distinction is the whole story of this page.

Epithalamin is a peptide extract of bovine pineal gland. It is the parent preparation, and it is the source of almost every longevity claim in this family. Given to fruit flies, mice and rats it raised mean lifespan by 11 to 31 percent. Given to elderly people alongside the thymic preparation thymalin in a 266-person observation over 6 to 8 years, the authors reported mortality falling 1.6 to 1.8 fold in the epithalamin group. In a separate 12-year study in elderly patients with coronary disease, the number of deaths in the epithalamine group was 28 percent lower than control.

Epitalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly, designed from epithalamin. It is a defined molecule, made in a lab, and it has its own separate literature.

Endoluten is neither. It is a retail capsule. No published analysis establishes that its contents match epithalamin, and a bovine gland extract is not a thing you can assume from a label. If you want the epithalamin numbers, you are quoting a Russian injectable gland extract from studies run by the group that developed it, not a capsule bought online.

Mechanism of action

The proposed mechanism is inherited rather than demonstrated. Epithalamin was reported to increase melatonin synthesis and secretion in rats and to inhibit free radical processes in rats and flies, and the authors of the 1998 lifespan paper suggested its antioxidant properties as the route to lifespan extension. A separate registered Russian pineal polypeptide preparation, Pineamin, raised urinary 6-sulfatoxymelatonin excretion 1.9-fold in 55 elderly patients in an uncontrolled before-and-after study, which is the most direct pineal-function measurement in the family. None of this work used Endoluten.

Human evidence

There is human data in this family, and none of it was generated with Endoluten. Two long observational studies of the injectable pineal extract epithalamin report large mortality reductions, and one small uncontrolled study of a third pineal preparation reports raised melatonin metabolite excretion.

  • 266 elderly and older persons, 6 to 8 years of follow-up, epithalamin given in the first 2 to 3 years: mortality reported 1.6 to 1.8 fold lower than control, and 2.5 fold lower when combined with thymalin.
  • 12-year study in elderly patients with coronary disease: 28 percent fewer deaths in the epithalamine group, cardiovascular mortality 2-fold lower.
  • 55 elderly patients given Pineamin, a separate registered Russian pineal preparation: urinary 6-sulfatoxymelatonin excretion rose 1.9-fold, uncontrolled before and after.
  • Zero indexed studies of Endoluten itself, under that name, in any species.
  • Zero registered clinical trials for any compound in this family on ClinicalTrials.gov.

What this does not tell you: Three limits, each sufficient on its own. First, the product mismatch: the studies used injectable bovine gland extracts and a registered Russian medicine, not an oral retail capsule, and no analysis has shown the capsule contains the same material. Second, the independence problem: the mortality studies are authored by the institute that developed the preparations and no outside group has replicated them. Third, the design: a 6 to 8 year observation with treatment only in the first 2 to 3 years and mortality as the endpoint is the kind of study where allocation and follow-up detail decide the result, and that detail is not recoverable from an English abstract.

Reading the research record

The reason this page separates three names so insistently is that the marketing does not. Epithalamin is a bovine pineal peptide extract and holds the lifespan and mortality data. Epitalon is the synthetic tetrapeptide Ala-Glu-Asp-Gly designed from it, with its own separate literature. Endoluten is a capsule sold at retail with zero indexed records under its name. Buying the third and quoting the first is the standard error in this category.

Everything in the Khavinson peptide bioregulator family shares one structural problem, and it is not a problem with the chemistry. Not one compound in the family has a registered clinical trial: ClinicalTrials.gov returns zero studies for epitalon, thymalin, cortexin, retinalamin, prostatilen and epithalamin alike, on a query shape that returns results for ordinary drugs. The literature sits almost entirely in three journals, Advances in Gerontology, Bulletin of Experimental Biology and Medicine and Vestnik Oftalmologii, most of it in Russian with English abstracts only, and most of it authored by the St. Petersburg Institute of Bioregulation and Gerontology or by Anisimov's group in the same city. No independent replication outside those groups has been located for any of the lifespan claims. Separately, no adverse event data appears in any of the abstracts retrieved for this family. The Russian literature in this area does not systematically report safety endpoints, and that absence should be read as missing data rather than as a clean safety record.

Nobody has tested it. The nearest measurement is a different preparation, Pineamin, raising a melatonin metabolite 1.9-fold in 55 elderly people with no control group. That is a biochemical change in a different product, not a sleep outcome in this one.

Is Endoluten safe?

Unknown. No adverse event data appears in any abstract retrieved across this entire family. The Russian literature does not systematically report safety endpoints, so read the silence as missing data.
