This is how I would explain it to a friend. The evidence, with the numbers, is further down the page.
N-Acetyl Semax Amidate is a nose spray sold for focus and a clearer head. It is regular Semax with two small chemical caps added to each end, which vendors say makes it last longer and hit harder. Not one study has tested this capped version in a person or an animal. Every bit of evidence belongs to plain Semax, and that is on the Semax page. This version is not approved anywhere.
What people take it for
- A stronger, longer lasting version of the Semax focus effect.
- Lifting brain fog.
- Better memory and faster learning.
- Mood and drive on low days.
What studies in people showed
Nothing has been tested with this version. No human study. No animal study. No study of how long it lasts in the body. The claim that it is stronger comes from the people selling it. What exists is for plain Semax. The human study on this page is a Russian trial of 110 stroke patients. They got 6,000 micrograms a day in two ten day courses. A brain growth protein called BDNF went up in their blood and they got better at daily tasks faster. There was no dummy group. Rats given plain Semax after a stroke had less brain damage. None of that was done with the capped version. Whether the caps change what it does, for better or worse, nobody knows.

- Some users say it lasts longer and feels stronger than plain Semax. That is what they are told to expect, so take it with salt.
- Brain fog lifts for a few hours.
The bad
- Nose burning, congestion or a runny nose from daily spraying.
- Headaches.
- Feeling wired or anxious, and trouble sleeping if it is used late in the day.
- Hair loss. This version is the one users blame most. Nobody has studied it, so nobody knows if it is real or why.
- Product quality is a coin flip. Batches that do nothing are a common complaint.
Where these come from: All of these are user reports from r/Nootropics, r/Peptides and nootropic write-ups. None of them come from a trial, because there is no trial of this version.
My bottom line
This is Semax with a vendor's promise stapled to it and zero data of its own. Read the Semax page for what the real thing has and has not shown. I would not pay extra for two chemical caps nobody has tested.

What is N-Acetyl Semax Amidate?
N-Acetyl Semax Amidate is Semax with both ends chemically capped. It is sold as a longer-lasting nose spray. No study of this form has been published, so you cannot assume Semax's Russian research applies to it.
Semax is a seven building block peptide approved in Russia and used there for stroke and memory problems. Sellers offer a version with both ends capped, claiming it lasts longer and works harder. No study of how this version behaves in the body, or whether it works, has been found. The changes could make it more stable, but that is unproven. It rides a sharp rise in searches for brain-boosting peptides.
How does N-Acetyl Semax Amidate work?
Assumed to work like Semax. That means acting on brain growth signals such as BDNF and on the brain's chemical messengers.
What have studies in people found about N-Acetyl Semax Amidate?
There is no evidence in people for N-Acetyl Semax Amidate. Searches of PubMed under each of its names find zero records of any kind, and no trial is registered. Nobody has published a study of this molecule in a person, an animal or a dish.
- Nothing published on the molecule itself: no study in people, no test of how the body handles it, no animal test, no lab test. It is sold as more stable and stronger than semax, with no published measurement of either.
- The parent peptide, semax, does have a record in people, in Russia. A 2018 Russian study followed 110 stroke patients in early or late rehab. Some were given semax at 6,000 micrograms a day in two ten-day courses. The treated group had higher blood levels of BDNF, a brain growth signal, and recovered daily-living skills better. That was plain semax, sprayed in the nose, in hospital, and reported in a Russian journal.
- Nobody has checked what is in the products. A 2020 forensic study of two products seized in Europe found semax and selank inside. It noted neither peptide has finished any clinical trial, and found both sold openly online as freeze-dried powder for injection and as nasal sprays. No lab test of a product labelled as this modified version was found.
What this does not tell you: Everything known about semax is known about a different molecule. The two caps added to this version, one at each end, are standard ways to slow a peptide's breakdown, so the stability claim makes chemical sense. But making sense is not the same as being measured. Nobody has published how long it lasts in the body, how much gets absorbed, whether it reaches the brain, or what dose matches plain semax. That cuts both ways. There is no proof it works, and no safety data either. No reported side effects here means nobody has looked, not that nothing happens.
The state of the research
Two different things are being mixed up under one product name. Separating them is most of what this page can usefully do.
The first is semax itself, which belongs to a real Russian medical tradition. It was made from a fragment of a stress hormone at the Institute of Molecular Genetics, registered as a medicine in Russia, and used there for stroke and memory problems for decades. There are Russian studies in people and many Russian animal studies on stroke. That work is not made up and not fringe. It is published, indexed and ordinary for its setting. It also has no repeat in the West, no large multi-centre study against a dummy outside Russia, and no review by any regulator outside Russia. Both halves of that are true, and neither is a verdict.
The second is the amidate version, and it is not part of that tradition at all. It is a seller's tweak of a Russian drug, brought into an English-language research-chemical market that wanted a product that sounded special. It borrows its trust from a parent it has never been compared with. Nobody has published on it because nobody has a reason to. A small change to a published recipe is not worth patenting. No regulator asks for data because it is sold as a research chemical. And no customer refuses to buy until the data exists. Every incentive points to making the claim, and none to testing it.
In practice, a buyer comparing N-Acetyl Semax Amidate with semax is comparing a studied compound with an unstudied one, and the gap is total, not partial. A reader who wants to weigh the Russian evidence is free to do so. They are weighing evidence about the other molecule.

Semax helped recovery after a stroke in a Russian trial. 110 patients were grouped by how soon rehab started and by whether they got semax. Those given it had more of a protein that supports nerve growth in their blood. They also got back more of their ability to handle everyday tasks. Keep one thing in mind. This was the original peptide, not the capped version sold online.
