Petrelintide (long-acting amylin analogue)

A once-weekly amylin analogue from Zealand and Roche being developed as a potentially better-tolerated obesity treatment, with positive Phase 2 results.

Overview

Petrelintide is a long-acting amylin analogue developed by Zealand Pharma and Roche as a stand-alone obesity therapy, on the idea that amylin-based drugs may be better tolerated than GLP-1 agonists. The Phase 2 ZUPREME-1 trial reported about 10.7% weight loss at 42 weeks, and it is planned to move to Phase 3 in the second half of 2026. It is investigational.

Mechanism of action

Agonizes amylin receptors to promote satiety and slow gastric emptying, used as a single-agent weight-loss approach.
