How does kisspeptin-54 work?
Kisspeptin-54 is the 54-residue product of the KISS1 gene. This site's profile says it binds KISS1R, formerly GPR54, on hypothalamic GnRH neurons, triggering release of endogenous GnRH, which drives pituitary luteinising hormone and follicle-stimulating hormone. That downstream LH response has been measured directly in human trials by subcutaneous, intravenous, and intranasal routes. Molecular weight is roughly 6 kilodaltons. A phase 2 trial of 60 women at high risk of ovarian hyperstimulation syndrome found oocyte maturation in 95%, and across 51 embryo transfers biochemical pregnancy, clinical pregnancy, and live birth rates were 63%, 53%, and 45%, with no moderate, severe, or critical OHSS. A second-dose trial in 62 women improved oocyte yield. Chronic administration causes tachyphylaxis. A 2025 randomised trial found reproductive hormones rose and anxiety did not change. There is no approved kisspeptin product. Those IVF numbers belong to kisspeptin-54, not kisspeptin-10. Chronic subcutaneous kisspeptin-54 caused testicular degeneration in adult male rats.
