How does FOXO4-DRI work?
FOXO4-DRI is a designed senolytic peptide. This site's profile says it disrupts the FOXO4-p53 interaction selectively in senescent cells, allowing p53 to initiate targeted apoptosis of those cells while sparing healthy cells. Half-life is listed as short. Molecular weight is about 5,200 daltons. It is not FDA approved. This site's review found no human clinical trial. The human mechanism has not been measured. The 2017 Cell paper reported restored fitness, fur density, and renal function in fast-aging XpdTTD/TTD mice and in naturally aged mice. A 2020 study found it induced p53 nuclear exclusion and apoptosis in senescent Leydig cells and improved testosterone secretion in aged mice.
