How does NMN work?
NMN is nicotinamide mononucleotide, a precursor the body converts into NAD+ through the salvage pathway. This site's profile lists molecular weight 334.2 daltons and half-life as short. It is not an approved drug. In 2022 the FDA excluded it from the US dietary-supplement definition because it had been authorized for investigation as a drug; US supplement status remains contested. Human findings on this site: a 10-week randomized trial in overweight or obese postmenopausal women with prediabetes found muscle insulin sensitivity rose versus placebo while other metabolic outcomes did not; a 60-day trial in 80 adults raised blood NAD and modestly improved six-minute walk distance, with HOMA-IR unchanged; 14 older men with diabetes and impaired physical performance, mean age 81.1, saw no significant grip or walking-speed separation over 24 weeks, Teijin funded; 30 overweight or obese adults on MIB-626, a microcrystalline NMN tablet, for 28 days lost 1.9 kg versus placebo, with diastolic pressure and LDL down, while muscle strength, aerobic capacity, and insulin sensitivity did not differ, maker funded. A blood NAD rise is not a clinical ageing result. No trial on this page has tested whether NMN slows ageing in a person. Twelve months of oral NMN in ageing mice improved insulin sensitivity, energy metabolism, physical activity, and eye function without obvious toxicity.
