What is the difference between semaglutide, tirzepatide, and retatrutide?
Each activates one more receptor than the last: semaglutide hits only GLP-1, tirzepatide adds GIP, and retatrutide adds glucagon on top of both. In their respective trials, average weight loss has scaled up the same way: roughly 15% for semaglutide's approved dose, up to 21% for tirzepatide, and up to 28% for retatrutide's Phase 3 dose, though these come from separate trials in different populations rather than one head to head study of all three.
