How does mazdutide work?

Mazdutide is a once-weekly dual agonist of the GLP-1 and glucagon receptors, based on the natural dual-acting hormone oxyntomodulin. Half-life on this site's profile is about 5 to 7 days. Molecular weight is about 4,800 daltons. Aliases on the profile are IBI362 and LY3305677. GLORY-1 randomized 610 Chinese adults: mean weight change of minus 10.1% at 4 milligrams and minus 12.6% at 6 milligrams versus plus 0.5% on placebo at week 32. A Phase 3 trial in 731 Chinese adults with type 2 diabetes found both mazdutide doses superior to dulaglutide 1.5 milligrams for HbA1c, with 3.8 to 5.8 percentage points more weight loss at 28 weeks. It was approved in China in June 2025 for obesity and in September 2025 for type 2 diabetes, the first GLP-1 and glucagon dual agonist approved anywhere. It is not FDA approved.
